[Tumor suppressor genes in human urological malignancies].

چکیده

はじめに 悪性腫瘍は今や日本人の死因の第1位 を占める疾患 であり, 過去4半 世紀の間に多くの進歩は見られたも のの, 現代でもその診断や治療は困難であることが多 い. 近年の分子生物学的な手法による遺伝子解析によ り, 癌の発生, 進展は特定の遺伝子群に起こるDNAの 損傷の蓄積 と考えられている. 化学発癌の考え方では 自然環境中に存在する発癌物質や, 自然界の放射線や, 紫外線によりこれらの遺伝子上のDNAの 損傷(=遺 伝子異常)が 引き起こされると考えられる. 癌化しつ つある細胞はしばしばこれらのDNAの 傷を修復する 機能を失っており, これらの異常が癌遺伝子や癌抑制 遺伝子のような特定の遺伝子群におこるがDNAの 損 傷が固定される結果, ある場合は直接, 細胞の増殖や 脱分化が進行し, (細胞の増殖を自動車にたとえれば自 動車のアクセルをかけることになり)そ の結果として ある種の遺伝子群は活性化された癌遺伝子として作用 する. また, 癌抑制遺伝子群は一般に増殖抑制の機能 を持っており, これが不活性化されることになり, 細 胞は増殖を抑制する機能を失ってしまい(自 動車のブ レーキの故障と同じ), 細胞増殖が無制限に進行する結 果となる. 一般的に多くのヒトの悪性腫瘍は多段階のステップ
